In 1977, Sanger developed a much simpler sequencing method, called “chain termination,” which is today known simply as Sanger sequencing. This technique took advantage of a different type of special nucleotide called a dideoxyribonucleotide, or ddNTP. ddNTPs lack one of the hydroxyl groups present on a normal dNTP, preventing the chemical reaction necessary to add another nucleotide and terminating DNA elongation.

He described the process as strangely therapeutic: “The further I progressed with my contingency plan, the more the effects of that trauma diminished.” He concluded, “My actions were not rational, but responses to extreme emotional trauma often are not rational.” He quoted Solzhenitsyn, as he’d done in the letter to me—“How can you expect a man who’s warm to understand a man who’s cold?”—and noted that he should have received therapy long ago.
